Aantal fracturen / botbreuken door osteoporose = botontkalking daalt door injecties met eigen gemodificeerde mesenchymale stamcellen met 94 procent jaarlijks in kleinschalige 6-jarige veiligheidsstudie met mensen met gevorderde osteoporose.
In een kleinschalige veiligheidsstudie met slechts 10 deelnemende patiënten - zie hier het studieprotocol - namen de onderzoekers eerst beenmerg af – specifiek autologe, uit beenmerg afkomstige mesenchymale stamcellen – uit de heup van elke deelnemer en kweekten ze miljoenen van deze stamcellen in het laboratorium.
Om ervoor te zorgen dat de gekweekte stamcellen zich goed aan de bloedvaten in het beenmerg zouden hechten of effectief het botweefsel zouden binnendringen, brachten de onderzoekers een suikerpatroon op het celoppervlak aan.
Dit deden ze door de gekweekte stamcellen in een vloeibare oplossing te plaatsen met een specifiek enzym / eiwit en een suikerdonormolecuul. Dit wordt in medische termen GDP-fucose genoemd. Door de GDP-fucose werden de cellen omgezet in HCELL waarbij de gekweekte stamcellen zich vasthechten aan receptoren aan de wanden van bloedvaten in het beenmerg.
Nadat de gekweekte stamcellen alle vereiste veiligheidstests hadden doorstaan, kregen de patiënten hun eigen aangepaste stamcellen toegediend via een infuus.
Artsen volgden de 10 deelnemende patiënten zes jaar met na twee jaar een controle onderzoek van de conditie van de botten door middel van scans, bloedonderzoek en botdichtheidsmetingen.
Gedurende de volledige studie follow-up van zes jaar daalde het aantal fracturen / botbreuken bijna volledig met 94 procent per jaar.
- Gedetailleerde 3D-botscans twee jaar na de injectie bevestigden dit beeld en lieten een duidelijke toename van de botdichtheid zien, tot diep in het botweefsel.
- Ook uit bloedonderzoek bleek een hoger niveau van markers voor botvorming; deze waarden bleven na slechts één infusie meer dan zes maanden verhoogd.
- Ook merkten de patiënten verschil in botpijn. Na 24 maanden was de botpijn met 34% afgenomen en waren de scores voor fysieke beperkingen met 30% verbeterd. Een heel goed resultaat dus.
